  • The pathogenesis and potential therapeutic stratiges of rheumatoid arthritis (RA)

    1. Mechanisms regulating the expression of potential pathogenic mediators in synovial fibroblasts and osteoblast including: signal transduction, transcriptional regulation, transcription factor/promoter interactions
    2. In vitro/in vivo correlation: A murine model of collagen-induced arthritis (CIA) were established. The role of pathogenic mediators in disease progression will be detected as listed. 
      a. the over-expressed or small hairpin (sh)RNA of the candidate genes will be constructed into lentivirus. These genes will be delivered to CIA rats via viral vectors.
      b. nano-technique for more efficient gene delivery will also be developed.
      c. transgenic mice targeting the potential mediators from I-1 will be established. 
    3. To substantiate the impact of the candidate molecules on RA development, the progression of CIA in groups“a”,“b”,“c”will be assessed by image analysis (X-ray or computer tomography) and immunohistochemistry. The results may help elucidating the therapeutic potentials of RA.

    Animal models of some commonly encountered dental diseases such as chronic apical periodontitis and biphosphonate-related osteonecrosis of the jaw (BRONJ) will be developed. The expressions of the mediators identified from I-1 will be detected.
